Grow Microorganism With Brain Heart Infusion Broth
Brain Heart Infusion Broth medium is a highly nutritive medium. It is used as a blood culture system for cultivating a wide variety of microorganisms. This culture medium complies with EN ISO 6888 and APHA standards. Brain Heart Infusion Broth Medium is a variation of Rosenow's original formulation, which included brain tissue pieces in dextrose broth. It is a non-selective, enriched broth medium that can be used to grow both fastidious and non-fastidious microorganisms. This medium will also allow aerobic microorganisms from clinical and non-clinical specimens to grow. Gentamicin and/or chloramphenicol are recommended for fungi-selective isolation.

Principle
Organic nitrogen, carbon, and vitamins are provided by the mixture of brain and heart infusions. The carbohydrate source is dextrose. To stimulate early growth, a low concentration of dextrose is used. The osmotic environment is maintained by sodium chloride. The buffering agent in this medium is sodium phosphate, which also helps neutralise the acids produced by dextrose utilisation and thus maintains viability.

How to Use Brain Heart Infusion Broth for Blood Culture?
Remove the plastic cap and disinfect the part of the rubber stopper that is now exposed.
Draw the patient’s blood with the sterile needle and syringe, and transfer the blood sample immediately into the culture bottle by puncturing the rubber stopper with the needle and injecting the blood.
Venting may be required for aerobic cultures but not for anaerobic cultures.
Incubate at 35–37 degrees Celsius for 18–48 hours, then for 7 days to confirm negative results.
