Plate Count Agar: A General Media For Sanitary Testing


Plate Count Agar, also known as Standard Methods Agar, is a microbiological growth medium that is commonly used to assess or monitor a sample's "total" or viable bacterial growth. Plate Count Agar is not a selective medium. It can be used to determine microorganism plate counts from a variety of samples, including foods, water, wastewater, and clinical samples. It is also used to count the number of microorganisms in other sanitary materials. This medium was formulated and described by Buchbinder et al. Plate Count Agar meets the requirements of ISO 4833-1 and 2:2013, as well as APHA. Principle



The medium consists of tryptone, which provides nitrogen, vitamins, minerals, and amino acids essential for the growth of microorganisms. Yeast extract is a source of vitamins, particularly the B group. Dextrose is a fermentable sugar, that provides carbon and energy. Agar acts as a solidifying agent.

Instruction For Use

  • In 1000 ml of distilled water, dissolve 23.50 grams.
  • Bring to a gentle boil while gently swirling to dissolve the medium.
  • Autoclave at 15 psi (121 °C) for 15 minutes to sterilise.
  • Allow cooling to 45-50 degrees Celsius.
  • Mix thoroughly and pour into sterile Petri dishes.
