Peptone special provides nitrogenous and carbonaceous compounds and other essential growth nutrients, while agar acts as a solidifying agent. How To Use TM Media’s Chromogenic UTI Agar
Dissolve 32.45 grams of TM Media’s Chromogenic UTI Agar in 1000 ml of distilled water.
Gently heat to boiling while swirling to dissolve the medium completely.
Sterilize by autoclaving at 15 psi (121°C) for 15 minutes.
Cool to 45–50 °C.
Pour into sterile Petri dishes after thoroughly mixing.
Growth Characteristics
The Enterococci form blue colonies when the enzyme ß-glucosidase cleaves one of the chromogenic substrates. The ß-D-galactosidase enzyme, which cleaves the other Chromogenic substrate, is what causes Escherichia coli to produce pink colonies. Coliforms create purple colonies by cleaving both the Chromogenic substrate. Tryptophan deaminase activity causes the colonies of Proteus spp., Morganella spp., and Providencia spp. to appear brown.
